چکیده

Influenzal encephalitis is frequently treated with oral antivirals, including amantadine (Symmetrel) for influenza A (if susceptible), rimantadine (Flumadine) for influenza A (if susceptible), and oseltamivir (Tamiflu) for influenza A and B; corticosteroids and immune globulin have also been tried. However, none of these agents has been proven to be effective for influenzal encephalitis. A combination of antiviral treatment, corticosteroids, and intravenous immune globulin has been suggested to reduce mortality due to influenzal acute necrotizing encephalopathy. There currently are no established therapies for West Nile virus encephalitis. Ribavirin (Rebetol), interferon, high-titer immune globulin, and corticosteroids have been used, and therapeutic trials are currently ongoing. No specific therapies have been proven to be effective for encephalitis due to other arboviruses or for rabies; successful use of coma-inducing therapy plus the antivirals ribavirin and amantadine was reported in one patient with rabies encephalitis. Corticosteroids, intravenous immune globulin, and plasmapheresis have been used for acute disseminated encephalomyelitis, but efficacy trials have not been performed.

PITPs as Targets for Selectively Interfering With Phosphoinositide Signaling in Cells

Sec14-like phosphatidylinositol transfer proteins (PITPs) integrate diverse territories of intracellular lipid metabolism with stimulated phosphatidylinositol-4-phosphate production and are discriminating portals for interrogating phosphoinositide signaling. Yet, neither Sec14-like PITPs nor PITPs in general have been exploited as targets for chemical inhibition for such purposes. Herein, we va...

Localization of Lipid Raft Proteins to the Plasma Membrane Is a Major Function of the Phospholipid Transfer Protein Sec14

The Sec14 protein domain is a conserved tertiary structure that binds hydrophobic ligands. The Sec14 protein from Saccharomyces cerevisiae is essential with studies of S. cerevisiae Sec14 cellular function facilitated by a sole temperature sensitive allele, sec14(ts). The sec14(ts) allele encodes a protein with a point mutation resulting in a single amino acid change, Sec14(G266D). In this stud...

Resurrection of a functional phosphatidylinositol transfer protein from a pseudo-Sec14 scaffold by directed evolution

Sec14-superfamily proteins integrate the lipid metabolome with phosphoinositide synthesis and signaling via primed presentation of phosphatidylinositol (PtdIns) to PtdIns kinases. Sec14 action as a PtdIns-presentation scaffold requires heterotypic exchange of phosphatidylcholine (PtdCho) for PtdIns, or vice versa, in a poorly understood progression of regulated conformational transitions. We id...

Differential regulation of Saccharomyces cerevisiae phospholipase D in sporulation and Sec14-independent secretion.

Saccharomyces cerevisiae Spo14, a phosphatidylcholine-specific, phosphatidylinositol (4,5) bisphosphate-activated phospholipase D (PLD), is essential for meiosis and spore formation. Spo14 is also required for secretion in the absence of the phosphatidylinositol/phosphatidylcholine transfer protein Sec14 (i.e., Sec14-independent secretion). In sporulating cells Spo14 is phosphorylated and reloc...
